Scientist/Principal Scientist - Stem Cell Engineering and Cell Line Development

We have a long history in the Diabetes Obesity and Complications TA of addressing the needs of individuals living with diabetes by providing breakthrough therapies that result in significant improvements in the lives of patients. Position Overview: We are seeking a highly skilled, passionate and motivated scientist to join our diabetes team. This position will play a key lab-based role in implementing iPSC gene editing capabilities at Lilly Seaport Center and will contribute to the development of the next generation of islet cell therapies. The principal scientist will also apply the iPSC platform to derive other cell types. Successful candidates will have experience with human iPSC cell culture, differentiation, banking, gene editing, molecular biology techniques and assay development. This position will report to Director – Islet Cell Therapy, Stem Cell Biology and Cell Engineering.

Key Responsibilities:

Establish lab infrastructure and iPSC platform capabilities, enabling and advancing iPSC gene editing and cell line development at new site. 

Optimize workflows for iPSC-editing, including guide RNA design, screening and delivery. 

Optimize and perform assays for on-/off-target analysis. 

Generate clonal gene edited iPSC lines and characterize via flow cytometry, DNA based assays and gene expression profiling, and sequencing techniques to advance candidates toward clinical stage.

Culture and execute routine directed differentiation of human iPSCs to islets and work collaboratively to assess drug substance attributes such as cell composition and function.

Evaluate and design mitigation strategies to ensure genomic stability of the iPSC lines and DS. Collaborate cross-functionally to support transfer of R&D assays to GMP.

Explore technologies to improve throughput and implement new workflows to automate processes.

Maintain accurate and detailed records of all experiments, synthesize results, prepare reports and present in meetings. Contribute to SOP development. 

Work seamlessly with management in executing workplans and meeting deadlines.

Basic Qualifications:

BS/MS in Stem Cell Biology, Cell Biology, Bioengineering, or a related field with 5+ years of relevant industry or academic experience in gene editing and cell therapy.

Hands-on experience with human iPSC culture, gene editing, and molecular biology techniques.

Additional Skills/Preferences:

Proficiency in gene editing, including design, optimization, and validation of guide RNAs. Hands-on experience with delivery methods for gene editing tools.

Strong understanding of genomic stability assessments and off-target analysis.

Excellent communication and interpersonal skills, with the ability to work effectively in a team-oriented environment.

Expertise in next-generation sequencing (NGS) techniques for detecting on-target and off-target editing events.

Experience in pancreatic islet directed differentiation is highly desirable

Hands-on experience in transitioning research-grade processes to GMP-grade is highly desirable.

Position located in Boston, MA. This is a full-time position requiring weekend cell culture responsibilities on a rotational basis.

Actual compensation will depend on a candidate’s education, experience, skills, and geographic location.  The anticipated wage for this position is

$69,000 - $179,300

Full-time equivalent employees also will be eligible for a company bonus (depending, in part, on company and individual performance).
